Hyperglycemia increase in oxidative stress results in diabetic complication. This study evaluated the anti-diabetie potential ofethanolic exiracis from root, leaf, green and ripe fruits of Morinda eitrifolia by determining their antioxidant and antihyperglycemic activities in vitro and in vho. The antioxidant activity was determined tlirough the measurement of DPPH radical scavenging actiYity in vitro. Mice were induced diabetes by using alloxan monoliydrate. The extracts of Morinda eitrifolia could suppress hyperglycemia in diabetic mice by the oral intake. Finally, the antioxidant effect of these plant extracts in the kidneys of experimental diabetic mice was obsen-ed through the total antioxidant status ITAS) in kidney of mice. The results showed that oral administration of ethanolic extracts (200 mg/kg x 2 times/ day) could make lower the glucose le^-el of blood as well as the commercial antidiabetic drug fglucofast). and reduced oxidative stress- mediated damage in diabetic kidneys. The results also proved that Morinda eitrifolia ethanolic extracts improved ihe DPPH radical scavenging activity in vitro.

EDTA (Merck), Fe(NH4)2(804)2 (Merck), acid acetic, alloxan monohydrate (AM) (Sigma), Trolox (Denmark), nudc mudi sinh ly. sodium benzoate, TBA (thiobarbituric acid) (Merck), thudc dilu tri benh tiiu dudng glueofast. diethyl ether va mgt sd hda chat khac.

2.2 Phirong p h i p

2.2.1 Trich eao edy Nhau bang dung moi ethanol

2.2.2 Khdo sdt hogt dong lam sgch goe tir do DPPH in vitro

Hoat ddng lam sach gdc tu do ciia cac cao ethanol tir cac bg phan ciia ciy Nhau dugc thuc hien theo quy trinh ciia Shirwaikar et al. (2006) cd dieu chinh nhu sau:

2.2.3 Khdo sdt khd ndng hg dirang huyet cua eao ethanol ede bo phgn cdy Nhau Chudt 9 tuin tudi khde manh dugc tiem dung dich alloxan monohydrate (AM) d ndng do 130 mg/kg trgng Iugng chudt de giy BTD. Sau khi chudt BTD dn dinh 7 ngay, kha n i n g ha dudng huyet cua cao ethanol la, re. trai xanh va trii chin ciy N h i u dugc xac dinh bang each cho chudt BTD udng thudc dieu tri B'TO glueofast (150 mg/kg trgng Iugng chudt) hoac cao ethanol tir cac bd phan cua ciy Nhau (200 mg/kg trgng Iugng chugt). Chudt BTD udng cao ethanol la, rl, trai xanh va trii chin 0,1 mlx2 lin/ngay trong 21 ngay dieu tri. Dudng huyet dugc xac dinh vao liic 8 — 9 gid sang trudc khi chudt dugc cho i n . Sau khi do dudng huylt 60 phiit, chudt dugc cho an va udng nudc binh thudng. Sau 21 ngay dugc dieu t n benh vdi cac cao chiet, chugt dugc cho nhin ddJ qua dem. thin dugc Iiy ra v i xu Iy cho cic thi nghiem tilp theo.

2.2.-f Khdo sdt su chong oxy hoa long so (Total Antioxidant Status assay (TAS)) in vivo

(superoxide). Neu trong thin cd hien dien cac chat chdng oxy hda thi se ket hgp vdi cac superoxide. Lugng superoxide cdn Iai se ket hgp vdi TBA de thanh lap TBA — RS (thiobarbituric acid reactive substances) dugc do d budc sdng 532 nm.

Quy trinh phan ung dugc thyc hien theo cac budc sau; 100 mg than chudt dugc tich ra nghien min vdi 200 ^l dung dich dem phosphate lanh ndng do 100 mM.

pH = 7,4. Sau khi Iy t i m trong 10 phut d 10.000 vdng/phiit, phin dich ndi d tren duoc sir dung cho viec khao sat hoat tinh chdng oxy hda cua cao chiet (Bhat et al.

2008).

TAS dugc x i c dinh theo phuong phap cua Koracevic et al. (2000) cd hieu chinh nhu sau: 10 ^L dich thin sau khi dong nhit (kidne\ homogenate) dugc pha loing vdi 490 |J.L dung dich dem sodium phosphate ndng do 100 mM. pH ^ 7,4 dugc cho vao hdn hgp gdm 0.5 mL dung dich sodium benzoate 10 mM vdi 0.2 mL Fe - EDTA (2 mL Fe - EDTA dugc pha tii 2 mM dung dich EDTA vdi 2 mM dung dich Fe(NH4)2(S04)2). Sau dd 0,2 mL H2O2 10 mM dugc cho vao hdn hgp phan ung. lie d i u va u d ST^C trong 60 phiit. Sau khi u, 1 mL acid acetic 2 0 % v i TBA (thiobarbituric acid) 0,8% trong NaOH dugc them vao dng nghiem. Hon hgp phan ung sau khi ii d 100°C trong 30 phiit dugc de ngudi d nhiet do phdng. Do h i p thu quang phd cua phin img dugc do d budc sdng 532 nm. TAS trong than dugc tinh toan tuang duang ndng do mM Trolox (Erejuwa et al, 2011).

3 KET QUA VA THAO LUAN

3.1 Sff an toan cua cao ethanol cac bo phan cay Nhau tren chu6t binh thiromg Dl dinh gii su an toan (khdng giy ddc tinh cip) ciia cao ethanol cac bd phan cay Nhau chudt nhit tring binh thudng dugc cho udng cao ethanol a nong dg 200 mg/kg trgng lugng. Kit qua vl sy thay ddi dudng huyet cua chugt dugc trinh bay trong hinh 1.

Theo kit qua hinh 1 cho thiy, dudng huylt cua chudt udng cic cao chiet va nhdm chudt ddi chimg thay ddi khdng khac biet cd y nghia thdng k& gifta cac nhom vi cac ngay udng cao chiit trong sudt qua trinh thir nghiem 21 ngiy. Nhu vay cao ethanol cac bd phin cay Nhiu khdng anh hudng din dudng huyet chudt binh thudng. Mat khac. trgng Iugng chugt sau 21 ngay udng cao ethanol cac bd phin ciy Nhau cung khdng khic biet cd y nghia thdng ke so vdi nhdm ddi chirng (ket qui khdng trinh bay).

3.3 K h a o sat ho^t tinh ch^ng oxy hoa ciia cao chiet cay Nhau

3.3.1 Khdo sdt khd ndng ldm sgch gdc tir do DPPH (2,2~Diphenyl~l-picrylhydrazyl) cua eao chiet tir ede bo phdn cdy Xhdu in vitro

Xac dinh kha ning chdng oxy hda bing phuang phap DPPH la phuang p h i p dan gian, nhanh chdng va it tdn kem. Trong nhimg nam gan day phuang phap niy Cling dugc sir dung d l djnh Iugng chat chdng oxy hda frong cic he thdng sinh hgc phuc tap (Prakash et al, 2000). DPPH la hgp chat cd miu tim dugc phat hien d budc sdng 517 nm. Khi cac dien di le cua cic gdc ty do DPPH kit hgp vdi hydro tir chat chdng oxy hda thi se hinh thanh nen DPPH-H liic nay m i u sic chuyin tir mau tim sang mau vang.
