ABSTRAK
Demam tifoid adalah penyakit infeksi akut pada saluran pencernaan yang disebabkan oleh bakteri Salmonella typhi. Peneliti di berbagai negara melaporkan adanya galur-galur Salmonella typhi yang resisten terhadap satu atau lebih antibiotik yang biasa digunakan untuk terapi penyakit tersebut. Penelitian ini dilakukan untuk mengetahui aktivitas antibakteri ekstrak dan fraksi umbi batang tumbuhan sarang semut (Myrmecodia pendens Merr. & L.M.Perry) terhadap Salmonella typhi NCTC786 BCC712. Penelitian dilakukan melalui tahap pengumpulan bahan dan determinasi tumbuhan, ekstraksi simplisia, penapisan fitokimia ekstrak, uji aktivitas antibakteri ekstrak, fraksinasi ekstrak, uji aktivitas antibakteri dan penentuan Konsentrasi Hambat Tumbuh Minimum (KHTM) serta Konsentrasi Bunuh Minimum (KBM) fraksi, penapisan fitokimia fraksi teraktif, dan penentuan profil Kromatografi Lapis Tipis (KLT) ekstrak dan fraksi teraktif. Hasil penelitian menunjukkan bahwa ekstrak etanol umbi batang tumbuhan sarang semut memiliki aktivitas terhadap Salmonella typhi NCTC786 BCC712. Ekstrak ini berupa serbuk berwarna coklat kemerahan dan berbau khas, dengan rendemen ekstrak 16,42% (b/b) dan kadar air 0,5 % (v/b). Fraksi air, fraksi etil asetat dan fraksi n-heksana dari ekstrak tersebut juga memiliki aktivitas terhadap bakteri uji yang sama, dengan fraksi teraktif berupa fraksi etil asetat (KBM 0,078% (b/v)). Fraksi ini berbentuk serbuk berwarna merah kecoklatan dan berbau khas, dengan rendemen sebesar 6,08% (b/b). Hasil penapisan fitokimia menunjukkan bahwa fraksi etil asetat umbi batang tumbuhan sarang semut mengandung metabolit dari golongan flavonoid, polifenol, tanin, dan kuinon.

Typhoid fever is a gastrointestinal infection disease that caused by Salmonella typhi. Researchers in several countries had reported strains of Salmonella typhi that is resistant to chloramfenicol, even resistant to some antibiotics (Multi Drug Resistant Salmonella typhi). The research was conducted to determine antibacterial activity of the ant plant stem tuber (Myrmecodia pendens Merr. & L.M. Perry) extract and fractions against Salmonella typhi NCTC786 BCC712. The steps of research were collection and determination of plant, simplisia extraction, phytochemical screening of extract, antibacterial activity test of extract and its fractions, determine Minimum Inhibitory Concentration (MIC) and Minimum Bactercidal Concentration (MBC) of fractions, phytochemical screening of the most active fraction, and analyze Thin Layer Chromatography (TLC) profile of extract and its most active fraction. The result showed that the ethanol extract of ant plant stem tuber has antibacterial activity against Salmonella typhi NCTC786 BCC712. The extract was brown-red colored with specific smell, yield of extract 16,42 % and containing 0,5% water. All of the fractions gave antibacteria activity with ethyl acetate fraction as the most active fraction (MBC 0,078%). This fraction was brown-red colored and specific smell with 6,08 % yield. Phytochemical screening result showed that the ethyl acetate fraction of ant plant stem tuber contains flavonoids, polyphenols, quinones, and tannins.
