UJI EFEK IMUNOMODULATOR EKSTRAK ETANOL DAUN
MAHKOTA DEWA (Phaleria macrocarpa (Scheff) Boerl.) PADA
MENCIT JANTAN
ABSTRAKImunomodulator adalah suatu senyawa yang dapat meningkatkan sistem imun. Salah satu tanaman yang memiliki aktivitas imunomodulator adalah daun Mahkota Dewa (Phaleria macrocarpa (Scheff) Boerl). Tujuan penelitian ini adalah untuk menguji efek imunomodulator ekstrak etanol daun mahkota dewa (EEDMD) pada mencit jantan.
Aktivitas imunomodulator ditentukan menggunakan metode bersihan karbon dengan mengukur laju eliminasi karbon, indeks fagositosis, dan indeks stimulasi. Kelompok perlakuan dibagi menjadi 5 kelompok, masing-masing terdiri dari 5 mencit. CMC-Na 1% (kontrol negatif), Imboost® (kontrol positif), EEDMD 50, 100, dan 200 mg/kg bb secara per oral selama 7 hari dan hari ke 8 diberikan suspensi karbon secara i.v. Pengambilan darah dilakukan pada menit ke 5, 10, 15 dan 20 lalu diukur absorbansinya menggunakan spektrofotometer UV-Vis. Mencit didislokasi di leher lalu diambil hati dan limpa. Dihitung laju eliminasi karbon, indeks fagositosis, dan indeks stimulasi.
Hasil penelitian menunjukkan bahwa pemberian dosis EEDMD 50, 100 dan 200 mg/kg bb dapat memberikan efek imunostimulan dengan peningkatan laju eliminasi karbon, indeks fagositosis, dan indeks stimulasi dibandingkan dengan CMC-Na 1% (p<0,05). EEDMD 200 mg/kg bb memiliki aktivitas fagositosis sebanding (p>0,05) dengan Imboost®.

EXTRACT IN MALE MICE
ABSTRACT
Immunomodulatory is a compound that can boost the immune system. One of the plants that have immunomodulatory activity is Mahkota dewa (Phaleria macrocarpa (Scheff) Boerl.) leaves. The purpose of this study was investigate the immunomodulatory effects of ethanol extract of Mahkota Dewa leaves (EEDMD) in male mice.
Immunomodulatory activity was determined using carbon clearance by measuring elimination rate of carbon, phagocytic index, and stimulation index. Treatment group was divided into 5 groups, each group consist of 5 mice. CMC-Na 1% (Negative control), Imboost (positive control), EEDMD 50,100, and 200 mg/kg bw were orally administered for 7 days and carbon suspension was given intravenous in day 8. Blood was taken at minute of 5, 10, 15 and 20 and then carbon absorbance was determined using spectrophotometer UV-Vis. The mice was dislocated in its neck and then was taken liver and limph. Elimination rate of carbon, phagocytic index and stimulation index were determined.
The results of research showed that the administration of dose of EEDMD 50, 100 and 200 mg/kg bw can give immunostimulatory effect by increasing elimination rate of carbon, phagocytic index, and stimulation index as compared to CMC-Na 1% (p<0.05). EEDMD 200 mg/kg bw have immunomodulatory activity comparable to Imboost®(p>0.05).
