TERHADAP RESPON HIPERSENSITIVITAS DAN TITER
ANTIBODI SEL IMUN MENCIT JANTAN
ABSTRAK
Mahkota dewa (Phaleria macrocarpa (Scheff) Boerl) merupakan tumbuhan yang daunnya memiliki aktivitas sistem imun yang bermanfaat untuk mempertahankan kondisi tubuh dari serangan penyakit. Kandungan kimia yang dapat meningkatkan sistem imun pada tumbuhan ini adalah alkaloid, flavonoid polifenol, tanin dan saponin. Tujuan dari penelitian ini adalah untuk melihat efek imunomodulator ekstrak etanol daun mahkota dewa terhadap respon hipersensitivitas dan titer antibodi sel imun mencit jantan.
Penelitian ini menggunakan 25 ekor mencit jantan. Mencit dibagi menjadi 5 kelompok, masing-masing kelompok terdiri dari 5 ekor mencit. Kelompok kontrol diberikan CMC-Na 1%, kontrol positif diberikan levamisol 25 mg/kg bb, dan kelompok yang diteliti diberi ekstrak etanol daun mahkota dewa dosis 50, 100, dan 200 mg/kg bb. Efek imunomodulator ekstrak dievaluasi berdasarkan volume pembengkakan kaki mencit dan hemaglutinasi titer antibodi sel imun.
Hasil penelitian diperoleh bahwa pemberian ekstrak etanol daun mahkota dewa dosis 50, 100, 200 mg/kg bb meningkatkan respon hipersensitivitas dan titer antibodi sel imun dibandingkan kontrol. Hanya dosis 200 mg/kg bb yang memberikan hasil yang tidak berbeda nyata dengan kontrol positif (p<0,05). Pemberian ekstrak etanol daun mahkota dewa dosis 200 mg/kg bb memberikan hasil yang lebih baik dibandingkan pada pemberian dosis 50 dan 100 mg/kg bb. Penelitian ini membuktikan bahwa ekstrak etanol daun mahokta dewa memiliki efek imunostimulator terhadap respon hipersensitivitas dan titer antibodi sel imun mencit jantan.

OF MALE MICE
ABSTRACT
Mahkota dewa (Phaleria macrocarpa (Scheff) Boerl) is a plant whose leaves have a beneficial immune system activity to maintain the condition of the body against disease. Chemical content that can boost the immune system in this plant are alkaloids, flavonoids, polyphenols, tannins and saponins. The objectivitas of this study was to evaluate the immmunomodulatory effect of ethanol extract of mahkota dewa leaves on hypersensitivity response and antibody titer of immune cells of male mice.
In this study 25 male mice were used. Mice were divided into 5 groups, each groups consist of 5 mice. Control was given 1% CMC-Na, positive control was given levamisole 25 mg/kg bw, and the studied group administered ethanol extract of mahkota dewa leaves at doses of 50, 100, and 200 mg/kb bw. Immunomodulatory effects of the extracts were evaluated based on the swelling of the feet of mice and hemagglutination titer antibody of immune cells.
The results showed that ethanol extract of mahkota dewa leaves at doses of 50, 100, 200 mg/kg bw enhanced hypersensitivity response and antibody titer immune cells as compared to the control. Only at dose of 200 mg/kg bw which gives results that are not significantly different from the positive control (p<0.05). Ethanol extract of mahkota dewa leaves at dose of 200 mg/kb bw gives better results as compared to the administration at dose of 50 and 100 mg/kb bw. The present study concluded that ethanol extract of mahkota dewa leaves showed immunostimulatory effects on hypersensitivity response and antibody titer immune cells of male mice.
