Abstract

The aim of this research was to obtain to get adaptive MSCs by an treatment in hypoxia precondition in viro culture. Ini this research, hypoxia precondition was to be given 3% O2 concentration and compared to those of normoxia culture in O2 > 20%.

Results showed that under 3% O2 concentration, CD105+ and CD45- by flowcytometri, immunocytokimia and immunofluorescence didn’t experience of change (undifferentiated). Meanwhile under > 20% O2 concentration, cells have experienced of change (not undifferentiated again), that was indicated with CD105+ become decrease and CD45- increase by flowcytometri, immunocytokimia and immunofluorescence.

Conclusion, in this research showed that hypoxia precondition with 3% O2 concentration very support MSCs to constantly adaptive before transplantated for disturbance of spermatogenesis process, because did’nt experience become progenitor cells was not expectation (still undifferentiated)

Abstract

Cellular transplantation using stem cells provides very promising solutions in the regeneration and repairment of cells that have experienced degeneration where recovery through medical or surgical intervension is impossible. However, the very low viability of transplanted stem cells limits the transplantation efficacy.

The aim of this research was to obtain viable bone marrow derived stem cells by an adaptation treatment in a low oxygen tensioned in vitro culture. Low oxygen tension adaptation was adjusted to the niche of the stem cells in vivo. In this study, in vitro culture of stem cells in 1% oxygen was compared to those of the conventional culture in 21 % oxygen.

Results showed that under 1% oxygen tension cell proliferation was slower with larger or rounded triangle shaped cells, and senescence or dead cells was low. Meanwhile under 21 % oxygen tension cell proliferation was two fold faster with flattened and slender cells, and senescence or dead cells was higher.

In conclusion, conventional in vitro culture under 21 % oxygen caused cell aging (senescence) and rapid cell death, therefore the transplanted cells were not viable.

Abstract

The aim of this research was to obtain to get MSCs that’s viable and self rennual for spermatogenesis process by an treatment in hypoxia precondition in vitro culture. Ini this research, hypoxia precondition was to be given 1% O2 concentration and compared to those of normoxia culture in 21% O2.

Results showed that : 1. under 1% O2 concentration, CD90+ and CD34- by flowcytometri didn’t experience of change (undifferentiated). Meanwhile under 21% O2

concentration, cells have experienced of change (not undifferentiated again), that was indicated with CD90+ become decrease and CD34- increase by flowcytometri; 2. under 1%

O2 concentration, by immunocytokimia and immunofluorescence, MSCs culture was expressed transcription factor, like as OCT4 and SOX2. Meanwhile under 21% O2

concentration, OCT4 and SOX2 as transcription factor (self rennewal function), have not expressed by immunocytokimia and immunofluorescence.

Conclusion, in this research showed that hypoxia precondition with 1% O2

concentration very support MSCs to constantly viable before transplantated for disturbance of spermatogenesis process, because still undifferentiated and always self rennewal.

Abstract

This research was conducted to learn of ovarian disfunction caused of malnutrition stress condition of female mice (Mus musculus) like premature ovarian failure (POF) in woman.

The methods used: The females of Mus musculus having malnutrition stress by fasting for 1, 2, 3, 4, and 5 days were given to drink ad libitum compared with controls fed a normal fixed 300-400 grams per day per mice and drinking water ad libitum. Further observations the celluler, by histological examination of the preparation of the ovary organ, based on counting the number of Subordinate follicles (follicles secondary and tertiary), dominant follicle and corpus luteum and histopathological examination, and examination of 5 times from estrus cycle by vaginal smears.

The study results show a significant difference between female mice made malnutrition stress compared with controls Mice with malnutrition stress led to inhibition of folikulogenesis developmental processes such as the formation of the decline is not even subordinate follicles or dominant or persistent corpus luteum formation (CLP) and histopathologic picture ovaries (hyperemia, oedema, congesti, and necrosis). In malnutririon stress serious condition can be cause of reproduction cycle disorders, such as not return of ofestrous again.

Abstract

The research was conducted in addition aims to scientifically prove the decrease in fertility of sperm cells of male mice Mus musculus experiencing malnutrition conditions, as well as to demonstrate the histopathologic picture of the seminiferous tubules were damaged in mice Mus musculus male from malnutrition. The methods used are: Mus musculus male is experiencing malnutri ie fasted for 3 days only given to drink ad libitum compared with controls fed a normal fixed 300-400 grams per day per mice and drinking water ad libitum.

We then test the fertility of sperm cells, sperm fertility rates obtained by bringing sperm and ovum cells in vitro is calculated based on the number of zygotes produced divided by the number of ova multiplied by 100%. The histopathologic examination of the seminiferous tubules of the testis is done by surgery and fabrication of histological preparations, and then observed microscopically at the damage occurs at 100 and 400 times. The results showed a significant difference between male mice created malnourished compared with controls, whereas in the treated mice showed a decrease in fertility malnutrition. Similarly, histopathologic observation of seminiferous tubules damage than the control. At the microscopic condition of malnutrition occurs in the seminiferous tubules swelling, bleeding and adhesin with the surrounding tissue and even degenerated tissue.

Abstract

This research was conducted with the long-term goal to improve the quality of livestock through scientific information on the bad effects and adverse effects of malnutrition for female animals. The specific target is to provide scientific information on the effects of malnutrition on reproductive dysfunction, infertiltas and sterility form the gonads (ovaries) in female mice as an animal model. The methods used are: females Mus musculus having malnutrition by fasting for 1, 2, 3, 4, and 5 days were given to drink ad libitum compared with controls fed a normal fixed 300-400 grams per day per mice and drinking water ad libitum. Further observations through several phases: 1. in vivo, based on an assessment of oestrous signs of the female, form female willingness to accept the stud marks (silent when riding), 2. the celluler, by histological examination of the preparation of the ovary organ, based on counting the number of Subordinate follicles (follicles secondary and tertiary), dominant follicle and corpus luteum and histopathological examination, and 3. in vitro by examination of vaginal smears 5 times of estrus cycle. Results from the study showed a significant difference between female mice made malnourished compared with controls, whereas in the treated mice led to a decline in malnutrition estrus signs (estrus) a decrease in the willingness of females accept males, dysfunction of folikulogenesis developmental processes such as the formation of the decline is not even subordinate follicles or dominant or persistent corpus luteum formation (CLP) and disorders of the reproductive cycle and histopathologic picture ovaries form of hyperemia, edema, and necrosis congesti.

SALMONELLOSIS INFECTION IN MOLTING PHASE WITH DIETARY FEED CARRIED OUT METHOD IN THE DUCKS

Abstract

The aims of this reseach is to give information to the farmers and people about the bad consequences of consuming the poultry products that use forced molting method. The research was carried out by examining Salmonella enteritidis bacteria in the duck feces after treatment.

The duck devided in five groups: the three groups were treated with dietary feed carried out method during 2, 4, and 6 days (forced moulting method), and another groups were given anti-molting (anti-prolactin) treatment by 100 μg/ml each hen single dose and control group (without treatment). Therefore the three groups were dietary feed carried out method compared with anti-prolactin method and control.

The result showed that forced moulting with dietary feed carried out during 2, 4, and 6 days decrease duck immune function. It was founded that Salmonella enteritidis in duck’s feces after treatment. The Salmonella enteritidis can be the causal factor of zoonosis for human. The Salmonella enteritidis was identificated with biochemical tests were done to various medium : TSIA, SIM, SCA, urea, and sugars.

The identification with TSIA medium of fermented carbohydrate, H2S and gas were positive. SIM medium identification was negative indole formation, positive H2S and positive motility. SCA was identified by blue medium. Urea Agar medium was uncreased urea production. Sugars agar medium was positive of glucose, lactose and sucrose, identified with acid production (yellow color) and negative of manitol and maltose, identified with alkaline production (no color change).

DETECTION OF ANTI-PROLACTIN RESIDUES AS A THERAPY FOR MOULTING DUCK

Abstract

The aim of this study was to know the effect of anti-prolactin with intra muscular injection on moulting phase of duck. Twenty female duck on moulting phase were subjected to this study. The ducks model devided evenly random into two factors, treatment dose and bleeding times. Four group of treatments dose are P1 (50 µg/ml), P2 (100 µg/ml), P3 (200 µg/ml) and five times of bleeding are 0 days or preimmunisation (control), 1 days, 2 days, 3 days and 4 days post immunisation. The blood samples were taken directly from Axillaris vein for anti-prolctin analyzed with indirect ELISA. The data were analyzed by univariate Anova and Tukey test using SPSS for windows program. The result showed that highly significant difference (p<0,01) for three dose of treatment. The bleeding time factor showed highly significant difference (p<0,01). Tukey tests showed that the highest value is at P3, for bleeding time, the highest Optical Density (OD) was found in one day after immunisation.

Furthermore, anti-prolactine doesn’t cause inflamation reaction, doesn’t leave residues and can be used for fastest moulting process.

UNNATURAL FORCED MOULTING IN THE LAYING HEN AS CAUSE OF ZOONOSIS FROM SALMONELLA ENTERITIDIS

Abstract

The aims of this reseach is to give information to the farmers and people about the bad consequences of consuming the poultry products that use unnatural forced moulting. The research was carried out by examining Salmonella enteritidis bacteria in the laying hens feces after treatment.

The laying hens devided in three groups and treatment with unnatural forced moulting, dietary feed carried out during 2, 3 and 4 days, and the another group given natural forced moulting treatment by 200 μg/ml each hen α Prl single dose compared with the group without dietary feed and α Prl.

The result showed that unnatural forced moulting with dietary feed during 2, 3 and 4 days decrease laying hen immune function. It was founded Salmonella enteritidis in laying hens feces after treatment. The Salmonella enteritidis can be the causal factor of zoonosis for human.

Abstract

The presence of increasing Vasoactive Intestinal Peptide (VIP ) of the poultry cause heat stress until now have much researched only interlocked with occurring of Prolactin hormone (PRL) increasing without specific explanation about how VIP can be synthesis by intestine. So that the examiner want to examine how VIP can be synthesis and how about the mechanism therefore can cause releasing of PRL. Research method of this examination is as follows: isolation, characterization, purification, specific test of VIP blood serum of chicken during realize heat stress with method Saturation Ammonium Sulphate (SAS) 50%, Western Blott, Electro Elusi, and Biuret method. Heat Stress is sabotaged by give temperature 40oC for 12 hours photo-period and then stabilized in temperature 24o-26oC. Inquiring specific Molecule Weight (MW) from VIP used SDS-PAGE 12% method. The common aim of this research is: to do identification an characterization of VIP isolated from layer chicken blood serum realize heat stress and to get heat stress biomarker at the laying chicken, therefore can be inquired the solution to handle it.

PREPARASI ANTIBODI POLIKLONAL ANTI-VIP

(VASOAKTIF INTESTINAL PEPTIDE) SEBAGAI ANTI HEAT SHOCK PADA UNGGAS PETELUR

Abstrak

Penelitian pada kasus Heat-shock selama ini hanya dikaitkan pada peningkatan PRH dari Hypothalamus yang akan mempengarhui Hypofisa anterior untuk mengeluarkan hormon PRL sebagai penyebab regresi ovarium sehingga ayam berhenti bertelur, padahal efek pertama yang muncul adalah dihasilkannya VIP dari Intestinal. Penelitian ini dilakukan untuk mempelajari lebih mendalam tentang preparasi antibodi poliklonal Anti-VIP pada hewan coba. Anti-VIP yang dipreparasi nantinya dapat digunakan untuk mengikat VIP yang tinggi dalam darah ayam yang mengalami heat shock sebelum terjadi peningkatan PRH (Prolaktin Releasing Hormon) dari Hypothalamus. Preparasi Anti-VIP dilakukan dengan menggunakan kambing jantan sebagai hewan coba. Produksi Anti-VIP dilakukan dengan cara penyuntikan pada kambing jantan intra cutan, dilakukan secara berulang (booster) dalam pelarut Complete Freund Adjuvant dan Incomplete Freund Adjuvant.

Metode preparasi antibodi poliklonal dilakukan dengan 2 cara berbeda, Metode I : booster dilakukan setiap 4 minggu sekali dan bleeding dilakukan 1 kali setiap minggunya, sedangkan Metode II : booster dilakukan tiap minggu dan bleeding dilakukan 2 kali setiap minggunya. Hasil dari kedua metode tersebut dibandingkan untuk mendapatkan metode terbaik dalam preparasi Anti-VIP. Isolat VIP yang digunakan pada penelitian ini adalah hasil isolasi dan karakterisasi serum ayam yang dikenai heat shock pada penelitian sebelumnya.

Hasil menunjukan bahwa metode terbaik untuk preprasi anti VIP adalah dengan metode I, dimana baik titer, kadar maupun jumlah volume anti VIP yang didapat lebih tinggi dibanding dengan metode II.

Abstrak

Penelitian ini dilakukan dengan tujuan jangka panjang untuk perbaikan mutu ternak melalui informasi ilmiah tentang efek buruk dan merugikan akibat malnutrisi bagi ternak jantan yang dipelihara. Adapun target khusus adalah memberikan informasi ilmiah tentang efek malnutrisi terhadap libido dan serta gambaran histopatologis dari testis mencit jantan sebagai hewan model.

Metode yang digunakan adalah : Mus musculus jantan dibuat mengalami malnutri yaitu dipuasakan selama 3 hari hanya diberi minum ad libitum dibandingkan dengan kontrol yang tetap diberi pakan secara normal 300-400 gram perhari per ekor dan air minum ad libitum. Selanjutnya dilakukan uji libido dengan cara mengumpulkan mencit jantan yang dibuat malnutrisi dengan mencit betina pubertas secara single matting. Uji libido dibedakan menjadi 4 karagori seperti : 1. Jumlah kawin per satuan waktu, yaitu berdasarkan frekuensi mencit jantan menaiki betinanya selama 60 menit; 2. Waktu reaksi atau waktu pada saat perkenalan hewan jantan dan hewan betina sampai saat kawin pertama kali, 3. Periode atau waktu diantara satu perkawinan dengan perkawinan berikutnya dan 4. Tingkat skor secara subjektif. Adapun Pemeriksaan histopatologis testis dilakukan dengan cara pembedahan dan pembuatan sediaannya kemudian diamati kerusakan yang terjadi.

Hasil menunjukkan perbedaan yang signifikan antara mencit jantan yang dibuat mengalami malnutrisi dibandingkan dengan kontrol, dimana pada mencit perlakuan malnutrisi menunjukan penurunan libido pada empat katagori uji libido yang diamati.

Demikian juga pengamatan pada testis secara histopatologis mengalami kerusakan dibandingkan kontrol, dimana secara mikroskopis testis mengalami odem, hemorrhagi, perlengketan dengan jaringan sekitar bahkan degenerasi.
