EFEK SITOTOKSIK EKSTRAK ETANOL DAUN AFRIKA (Vernonia amygdalina Del.) TERHADAP SEL HELA DAN VERO
ABSTRAK
Kanker serviks merupakan salah satu kanker penyebab kedua kematian wanita di Indonesia setelah kanker payudara. Kanker adalah pertumbuhan sel-sel tubuh yang tidak terkendali atau abnormal. Daun Afrika (Vernonia amygdalina Del.) banyak digunakan sebagai antioksidan, antidiabetes, antimutagenik dan belum ada data mengenai efek sitotoksiknya. Oleh karena itu dilakukan penelitian efek sitotoksik daun Afrika terhadap sel HeLa dan Vero untuk mengetahui potensinya sebagai antikanker. Tujuan penelitian ini adalah untuk menentukan nilai IC50 ekstrak etanol daun Afrika terhadap sel HeLa dan Vero dan untuk mengetahui tingkat keselektifan ekstrak etanol daun Afrika.
Uji aktivitas sitotoksik dilakukan dengan metode MTT [3-(4,5-dimetil- tiazol-2-il)-2,5-difenil tetrazolium bromida]. Sel HeLa dan Vero dibiakkan dalam media kultur RPMI pada 96-well plate kemudian diberi ekstrak etanol daun Afrika dengan seri konsentrasi 500 µg/ml, 250 µg/ml, 125 µg/ml, 62,5 µg/ml, 31,25 µg/ml, 15,625 µg/ml. Hasil pengujian dibaca dengan ELISA reader pada panjang gelombang 595 nm.
Hasil pengujian aktivitas sitotoksik ekstrak etanol daun Afrika terhadap sel kanker HeLa menunjukkan nilai IC50 sebesar 61,357 µg/ml dan Vero sebesar 79,561 µg/ml. Hal ini membuktikan bahwa ekstrak etanol daun Afrika memiliki efek sitotoksik yang poten terhadap sel kanker serviks HeLa.

CYTOTOXIC EFFECT OF ETHANOL EXTRACT OF LEAVES AFRICAN (Vernonia amygdalina Del.) ON HELA AND VERO CELLS
ABSTRACT
Cervical cancer is the second leading cause of cancer death in Indonesian after breast cancer. Cancer is cells uncontrolled or abnormal. Africa leaves (Vernonia amygdalina Del.) are widely used as an antioxidant, antidiabetic, antimutagenic and there are no data regarding the cytotoxic effects. It have to do study the cytotoxic effects of African leaf on HeLa and Vero cells to determine its potential as an anticancer. The purpose of this study was to determine the IC50 value of ethanol extract of leaves African against HeLa cell and Vero cells and to determine the degree of selectivity of ethanol extract of leaves of African.
Cytotoxic activity test was conducted using the MTT [3-(4,5- dimetil- tiazol-2-yl)-2,5 difeniltetrazolium bromide]. HeLa and Vero cells cultured in RPMI culture medium in 96-well plate and then given the ethanol extract of leaves Africa with a series of concentration 500 µg/ml, 250 µg/ml, 125 µg/ml, 62.5 µg/ml, 31.25 µ g/ml, 15.625 µg/ml. The test result is read with an ELISA reader at a wavelength of 595 nm .
Test results cytotoxic activity of ethanol extract of leaves African against HeLa cancer cells showed IC50 value of 61.357 µg/ml and Vero at 79.561 µg/ml. This proves that the ethanol extract of the leaves of Africa has potent cytotoxic effects against cervical cancer HeLa cells .
