Chuan bj mau thir

Djch chiet nu-dc thu du-oc bing phu-ong phdp sic vdi dung mdi Id nu-dc (dun sdi trong 1 h, Idp lai 3 lin). Gdp djch chiet nu-dc, bay hoi dung mdi den dang cao Idng 1:1. Djch chiit con thu du-oc bing phu-ong phdp chiit hoi lu-u vdi dung mdi ethanol 90° trong 1 h, Idp Igi 3 lin. Bay hoi dung mdi d i n c i n . C i n du-oc hda tan lai vdo nu-dc (ehda CMC 0,5%). Miu thd (djch chiet nu-dc, djch chiit con) du-oc chuan bj vdi lieu phu hop de cho chudt thi nghidm u6ng.

Thuoc, hda chat

Thuoc doi chieu: diphenhydramin cua Cdng ty Du-gc phim VFnh Phuc, aminophyllin cua hang Gedeon Richter (Hunggari), histamin dihydrochlorid cua Biomedicals (Phdp), thiopental cua RotexMedieal (Ode), xylazin cua Keprc) (Hd lan).

Dgng vat thi nghidm

Chudt lang, ca 2 giing, trong lu-ong td 350- 400g, khoe manh do Hoc vidn Qudn Y cung cip. Chudt dyoc nudi 6n djnh trong diiu ki0n phdng thi nghidm 2 tuin tru-dc khi thyc hi$n nghidn cdu, dyp-c nudi dydng bing thdc Sn tidu chuin, nydc u6ng ty do.

IVIay moc, thiet bj, dung cy

Thiit bj nghidn cdu co thit p h i quan g i m 3 bd phdn: mdy thdng khi hd trp- (Rodent Ventilator, model 7025), bO ghi vS khuyich dai tin hidu co thit (Bronchospasm transducer model 7020), mdy ghi d i u dd 1 kdnh (Uni record model 7050) cua hdng Ugo Basile (Y).

Sau 15 phut (vdi cac Id doi chieu) vd sau 1,5 gid (vdi cac Id thd uong cdc djch chiit cua x i u ho), tidm tac nhdn gdy eo thit histamin vdi lieu da du-oc lya chon. Ghi ddp dng eo thit lidn tyc trong vdng 20 phut. Kit qua du-oc bieu didn theo ty Id phin trdm eo thit so vdi eo thit eye dai tai cac thdi diim 5, 10, 15 vd 20 phut sau khi tidm histamin. Chung tdi dd trien khai thdnh cdng md hinh gdy CO thit CO tron phi quan tgi cho trdn chudt lang de ddnh gid tdc dyng cua thuoc (ho$c du-oc lidu) theo hu-dng dieu trj hen phi quan. Lieu 5 ng/kg histamin Idm tdng cd y nghTa ddp dng co thit vd duy tri ddp dng ndy trong suit thdi gian theo ddi.

Djch chiit nu-dc vd dich chiit con cua xiu ho vdi liiu 5,6 g/kg cd tdc dyng dc che ddp dng co thit phi quan trdn md hinh. Tdc dyng dc che cua djch chiit nu-dc vd djch chiet c i n cua xiu ho tirong du-ong vdi tdc dyng cua diphenhydramin (lieu 2 mg/kg) vd aminophyllin (lieu 6 mg/kg).

Summary

7776 standard procedure of respiratory phanvacology for evaluating bronchospasmolytic activity in anesthetized guinea pigs (modified Konzett-Rosseler's) which had not been applied extensively in Vietnam to evaluate bn:)nchospasmolytic effects of medicinal plants was used in this study to estimate the bnxhospasmolytic effects of the aqueous and ethanol extracts of Mimosa pudica L, with histamine (5^ig/kg, IV) as spasmogen. The plant aqueous and ethanol extracts exhibited the desirable effect at dose of 5.6 g/kg, po, significantly reducing bronchospasm as compared to the control This bronchospasmolytic activity was comparable to that of diphenhydramine (2 mg/kg, IV) and aminophyllin (6 mg/kg, IV). Further evaluations should be required for this promising medicinal plant in treating asthma.

Summary

A preliminary research about the effect of drug quality through period of store by temperature and humidity at different geographic areas was earned on three kinds of pharmaceuticals including Aspirin pH8 entenc-coated tablets, Vitamin C film-coated tablets and Amoxicillin capsules of different companies preserved at Thuan An coastal area, Nam Dong mountainous region, flat country of Hue Province and Hue Drug Quality ContnDl Centre. Result showed a considerable decrease of drug quality affected not only by high temperature and humidity in stored period at different areas but also by the stability of each active compound and each produced formula in one kind of different

drugs. The decrease of drug quality happened earliest and strongest at Thuan An, and then Nam Dong, where were sustained a severe climate condition yearly. Especially, effect of humidity factor on the drug decomposition was more serious than temperature. Almost of drugs stored at these areas was less quality after three to five months, presented clearly by the change of physiochemical properties such as appearance and active compound concentration. The decrease of drug quality was also happened at Hue city but in gentle degree, while all drugs were still in good quality under the standard store condition at Hue Drug Quality Control Centre (2^C. 60%) This research showed an importance of the temperature and humidity control during the store and delivery periods to maintain the drug quality sufficiently for treatment.
