S U M M A R Y

The Realtime-PCR using EvaGreen as an intercalator dye was performed to detect and identify human malaria Plasmodium spp. The result showed that Realtime-PCR is a reliable mediod for parasite detection and be able to differenciatc Plasmodium species based on melting temperanire (Tm). In diis study, the specimens inchiding 74 of P.faldpanim, 39 of P.vivax and 5 of P.malariae, »*ich were confirmed by Nested PCR, were analyzed, and ftie average melting tOT^waftires of each species was identified as follow: 75^4 ± 0,46; 78,55 ± 0,51 and 74,16 ± 0,08, respectively. The result also revealed that the sensitivity, specificity and accuracy of realtime PCR are higher compared to Nested PCR, Eiiber Realtime PCR or Nested PCR is alternative method for clinical malaria diagnosis.

However realtime-PCR reveals more advantagous such as: the result can be observed on the machine during running assay, short time requirement (ie.about 3 hours to complete); no need for electn^horesis. Hie parasites can be quantified, which allow to monitor the eflectiveness and the reqxinse of antimalaria d m ^ to parasites during treatment in order to give ^proriate therapies.
