PEMBUATAN SELULOSA MIKROKRISTAL DAN EKSTRAK ETANOL DAUN NENAS ( Ananas comosus (L.) Merr.) YANG DIFORMULASIKAN
MENJADI SEDIAAN TABLET
ABSTRAK
Latar Belakang: Daun nanas (Ananas comosus (L). Merr.) termasuk limbah yang mengandung selulosa yang dapatdibuat menjadi bentuk kristal. Selulosa mikrokristal merupakan bahan tambahan yangsering digunakan dalam pembuatan tablet dengan metode cetak langsung.
Tujuan:Penelitian ini bertujuan untuk membuat dan mengkarakterisasi selulosa mikrokristal dan ekstrak etanol dari daun nanas yang diformulasikan menjadi sediaan tablet.
Metode: Serbuk simplisia diekstraksi dengan etanol 80%, maserat yang diperoleh diuapkan dengan penguap putar. Selulosa mikrokristal daun nanas diperoleh dengan cara memanaskan daun nanas dengan NaOH 4%, dinetralkan kemudian diputihkan dengan NaOCl 2,5%, dinetralkan, dipanaskan dengan NaOH 17,5% dan dinetralkan. Hasil yang diperoleh adalah alfa selulosa yang kemudian dihidrolisis dengan HCl 2,5 N untuk mendapatkan selulosa mikrokristal, yang kemudian dikarakterisasi, dibandingkan dengan Avicel PH 102. Selulosa mikrokristal yang diperoleh dijadikan sebagai bahan eksipien dalam pembuatan tablet secara cetak langsung.
Hasil: Selulosa mikrokristal daun nanasmempunyai rendemen sebesar 29,49% dan hasil karakterisasi selulosa mikrokristal daun nanas dan Avicel PH 102 yang diperoleh secara berturut-turut adalah untuk uji organoleptik berupa serbuk berwarna putih, tidak berbau dan tidak berasa; pH 6,9 dan 6,5; susut pengeringan 5,19 dan 4,75%; kadar abu total 0,35 dan 0,01%; kelarutan zat dalam air 0,023 dan 0,08%; bobot jenis nyata 0,570 dan 0,41 g/cm3; bobot jenis mampat 0,668 dan 0,48 g/cm3; bobot jenis benar 1,43 dan 1,46 g/cm3; indeks Hausner 1,17 dan 1,15; indeks kompresibilitas 17,31 dan 15,86%; porositas; 60,4 dan 71,51%. Analisis FT-IR selulosa mikrokristal daun nanasdan Avicel PH 102 mempunyai spektrum yang sama dan dengan menggunakan alat pemindai elektron menunjukkan bentuk tidak beraturan dengan tekstur permukaan yang tidak rata.Hasil evaluasi tablet ekstrak etanol daun nanas dengan bahan pengisi selulosa mikrokristal dan Avicel PH 102untuk uji kekerasan diperoleh4,00 dan 4,62 kg; uji friabilitas sebesar 0,77 dan 0,68%; uji waktu hancurnya 3 menit 18 detik dan 2 menit 34 detik.
Kesimpulan: Berdasarkan hasil penelitian disimpulkan bahwa selulosa mikrokristal dapat dibuat dari daun nenas dan dapat diformulasikan menjadi sediaan tablet yang memenuhi persyaratan.

PREPARATIONOF MICROCRYSTALLINE CELLULOSE AND ETHANOL EXTRACT THE LEAVES OF PINEAPPLE (Ananas comosus
(L.) Merr.) ARE FORMULATED INTO DOSAGE TABLET
ABSTRACT
Background: The leaves of pineapple (Ananas comosus(L.) Merr.) including waste containing cellulose, can be isolate and made into a crystalline form. Microcrystalline cellulose is an additional ingredient that is often use in the manufacture of tablets by direct compression methods.
Purpose: The aims of this study was isolated and characterize the microcrystalline cellulose and ethanol extract from pineapple leaves was formulated into dosage tablet.
Methods: Dried material powder was extracted with 80% ethanol, macerate was consentrated with a rotary evaporator. Pineapple leaf microcrystalline cellulose was obtained by heating the pineapple leaves with 4% NaOH, NaOCl bleached with 2.5% and 17.5% NaOH was heated and result alpha cellulose which hydrolyzed with 2.5 N HCl to obtain microcrystalline cellulose, which was characterized, compared with Avicel PH 102. The microcrystalline cellulose was formulated into a tablet by a direct compression method.
Results: Microcrystalline cellulose from pineapple leaf yielded 29.49% and the result of microcrystalline cellulose pineapple leaves and Avicel PH 102 were obtained respectively for organoleptic tests in the form of white powder, odorless and tasteless; pH 6.9 and 6.5; drying shrinkage of 5.19 and 4.75%; total ash content of 0.35 and 0.01%; solubility in water of 0.023 and 0.08%; apparent specific gravity 0.570 and 0.41 g / cm3; incompressible specific gravity 0.668 and 0.48 g / cm3; true specific gravity of 1.43 and 1.46 g / cm3; Hausner index of 1.17 and 1.15; compressibility index of 17.31 and 15.86%; porosity; 60.4 and 71.51%. FT-IR analysis pineapple leaf and microcrystalline cellulose Avicel PH 102 had a similar spectrum and a Scanning Electron Microscopy (SEM) showed irregular shapes with uneven surface texture. The results of the evaluation of the ethanol extract from pineapple leaf tablets with excipients of microcrystalline cellulose and Avicel PH 102 for hardness test obtained 4.00 and 4.62 kg; friability test of 0.77 and 0.68%; time trials destruction of 3 minutes 18 seconds and 2 minutes 34 seconds.
Conclusion: Based on the result of the study microcrystalline cellulose can be made from the leaves pineapple and can be formulated to tablet preparation which meet the requirements.
