Development of a serological technique for the titration of antibodies for polioviruses in
children
Ehab Husain Mattar Abstract
Poliomyelitis (polio) is a highly infectious disease caused by a virus. It invades the nervous system, and can cause total paralysis in a matter of hours. It can strike at any age, but affects mainly children under three (over 50% of all cases). The virus enters the body through the mouth and multiplies in the intestine.
In the present study host cell susceptibility to infection with attenuated polioviruses strains was reviewed with 6 types of cell lines, Vero, RD, Hep2, A549, MDCK and HSF. Infectivity was measured by staining with trypan blue dye and microscopic examination. The results revealed that Vero and RD cells were highly susceptible for attenuated poliovirus infection. No cytopathic effects were observed on other cell types during the incubation period of ten days. Immune response against oral
poliovirus vaccine was also measured between vaccinated children and young adults between the age of one month to fifteen years. It was noticed that children of 1-2 months of age gained a protective immunity against polioviruses from their mothers.
The responsiveness against poliovirus vaccine was varied much between children at different ages. It was noticed that some of the studied children have no protective levels of antibodies which represents a high risk factor if they are exposed to wild polioviruses.
