In plant, auxin plays cntical roles in many aspects of plant growth anti cieveiopment. Aitliougii a number of auxin biosynthetic pathways have been identified. Recently AlYUCCAfJ gene belonging to the YUCCA family, encoding putative flavm monoox>'genase enzymes has been reported playing cnUca! roles of plant gtowtlt and development. Overexpressing .il] I CC.Uigene mcreases endogenous lAA levels response to environment stresses .is drought-tolerance. By PCR. we amplified coding sequence (1.25 kb) oi AiyUCCA6 gene from .Arabidopsis thahana and digested with Soft and SpA. AIYUCCA6 gene was then inserted in between .\\>!\ and SpA sites of pBluescript vector that cam'ing LTBQH promoter. Results of cloning were confirmed by enzyme di.gcsuon and sequencing. pliBQlt ..^iMJCCAG construct was cut by Asd and S/jd enz\'iiies and transferred to expression vector pERS at the same posiUon of those enzymes and confirmed by colony PCR and enzyme digesuon. Consequently. pER8-pUBQ14:AtYUCCA6 was transformed into .^grobaderium ti\'3101 sU-am for plant U-ansformation.
