Benh bac la do vi khuan Xanthomonas oryzae pv oryzae (Koo) gay ra la mot benh phd biln tren cay lua d mpi noi tren thi gidi. Thi nghiem dugc thuc hien blng phuong phIp chi thi phan tii vl danh gia kieu hinh thong qua phan ling vdi chi thi 13 ndi benh bac II tai elc tinh DBSCL. Cac gidng khang se dupe tiep tuc chpn Ipc dl dua ra sin xuat hoac phuc vu cong tic nghien ciiu tilp theo. Cac chi thi phan tii dugc diing nhu: RM 122, RM 13; da tich dupe 16 gidng mang gien xa- 511: OM 4495, OM 2517, OM 2518, OM 2514, CM 5239, OM 5240, OM 2490, OMCS 2000, OM 1490, OM 5636, OM 3408, OM 3238, OM 4096, OM 3010, OM 2403, OM 2280. Su dung chi thi PB7-PB-8 da tieh dupe chin gidng mang gien Xa-21 nhu: OM 4488, OM 5637, OM 4214, OM 4218, OM 4096, OM 6778, OM 5886, OM 3536 va OM 10011; gidng OM 4498 mang gien xarl3, ba gidng mang gien Xa-10^\i\x. OM 4872, DS 2002, OM 4285; bdn gidng mang gien Xa-i'nhu: OM 3837, OM 4213, OM 3834, MNR 5; mang gien xa-1 cd d c gidng: OM 1238, OM 3432, OM 2760, OM 4225, OM 5756; hai gidng mang gien Xa-14\k OM 8928 va OM 3243; vl gidng MNR 1 mang gien Xa-4.

LDATVANDE D. VAT LIEU VA PHUONG PHAP

B$nh bac II do vi khuan Xanthomonas oryzae pv oryzae (Xoo) gay ra II mot benh phd biln tren cay ilia d mpi noi tren thi gidi. Tai ddng blng song Ciiu Long (DBSCL) vi khuan Xoo cd thi llm gilm hon 30% nang suit liia. Cd 11 ndi vi khuan dugc xlc dinh tai Philippines, hai ndi tai An Dp, mdt ndi tai Vien Liia DBSCL ( Viet Nam) vdi d c gien khIc nhau (Lang va ctv, 2005). Ngoai ra cd 24 gien khie lien quan den d c ndi ciia Xoo vl da dugc phan nhdm kieu gien. Viec chpn Ipc d c ddng khang llm vat lieu de phuc vu cho cong tic chpn tao gidng liia la dieu rat khd. Nhilu kit qui nghien ciiu da cho thIy nlu chi dinh gia kilu hinh don doc se khong chinh xlc.

Chpn gidng blng chi thi (marker) phan tu se giiip nit ngin thdi gian chpn lua gidng thich hpp cho timg gien mong mudn. Bai bao nay trinh biy kit qua nghien ciiu thong qua phan tich kieu hinh va kilu gien ddi vdi phan ling gien benh bac la ciia cac gidng ilia mua ti-ong ngan hIng gien cua Vien Liia DBSCL, qua dd tim ra cac gidng khang voi cac noi gay benh bac la khac nhau.

Liia sau khi cay 30 ngay dugc cit la (khoang 3-5 cm) de lay nhilm benh, ngudn benh sau lay nhilm dugc bao trong giay bdng md vl ghi nhan d n than vdi timg nghiem thiic.

Mot tuan sau khi lay nhilm benh thi cat la de do chilu dai vet benh va phan nhdm khang nhilm theo he thdng danh gia ciia Vien Nghien ciiu Liia Qudc tl:

Chilu dai vet benh d<5 cm dupe dinh gia la khang (R), d>5 cm dupe danh gia la nhilm (S). Cac ddng vi khuan da phan lap dugc d y chuyen trong tu cay v6 tning tren dia thach (agar) chiia moi trudng nuoi d y vi khuan va dupe u trong phdng nuoi d y trong 3 ngay. Sau dd chuyin 1 khuan lac vao binh tam giac cd chiia 100 ml nude cat v6 tning. Cho binh tam giac len may lie, lie deu trong 30 phiit liic nay ndng dp vi khuan dat 10* tl bao/ml dugc dimg de lay nhilm len la Ilia. Dung dich dupe pha tuoi mdi ngay.

- Danh gia 6 microsatellite markers (chi thi ve tinh nhd) va 1 STS markers (chi thi vi tri chudi danh dau) cd tinh da hinh cho gien khang benh bac la lien quan.

- Xet nghiem STS (sequence tagged site - vi tri chudi danh dau) marker theo Lang, 2002.

III. KET QUA VA THAO LUAN

1. Danh gia d c gidng chi thi khang vdi ndi b^nh bac II tai DBSCL

Cac gien khang bac la dupe thilt kl trong bp gidng chi thi khac nhau (differentials) tai Vien

Blng 1. DInh gil 13 ndi b#nh bac la tr6n

Nghien ciiu Liia Qudc tl dupe su dung lam ca sd cho viec xlc dinh cac ndi dang hien dien tai DBSCL. Tiln hanh chiing 13 ndi tren nhiing gidng chi thi cd gien khing (blng 1). Trong thi nghiem danh gil kilu hinh, khao sat cac gidng chi thi (ddi chiing) phan ling vdi 13 ndi vi khuan gay benh bac la liia tren cac gidng Ilia chuan khang (Te tep) va gidng chuan nhilm (IR 24). Kit qui trinh bIy d blng 1 cho thay, gidng IRBB2 thi hien phan ling khang vdi 10 ndi, ngoai tni ndi sd 5 (Long An), ndi 3 (Can Tha), ndi 9 (Vinh Long) va ndi 10 (Bac lieu). Tuong ti gidng IRBB21 cho phIn ling khing cao vdi 9 ndi vi khuan gay benh, chilm ti le 70% sd ndi khing, ngoai tni ndi 5 (Long An) va ndi 8 (Tri Vinh) va ndi 12 (Ca Mau).

Kit qua nay cung phii hpp voi kit qui trude day ciia Dang Quang Hlo vl Nguyin Thi Lang (2006). Ngupc lai d hai gidng IRBB4 vl IRBB8 cho phIn ling khang thap, chi khing dupe 4 ndi vi khuan (30,3%), thi hien phan img nhilm cao vdi 9 ndi vi khuan, chilm ti le 64%. Hai gidng IRBB5 va Nang Tay Dilm khang dupe 7 ndi. Cdn lai cac gidng khIc deu cd phIn ling khang tii 5-6 ndi nhu la gidng IRBBl, IRBBIO, IRBB 7...

Phan img khang ciia timg ndi vi khuan ciing dupe ghi nhan d cac gidng ddi chiing cd ti le khang cao voi vi khuan bac II. Cu thi ndi 4 (Ddng Thap) khang voi 12 gidng (chilm 90%), ndi 13 (Hau Giang) khang vdi 10 gidng chilm 70%. Da sd gidng nhilm voi ndi sd 5 , sd 8 va sd 9.

2. Tim gien khang b$nh bac II h-ong d c gidng liia cao san

Tiln hinh khIo sat phIn ting khang ciia 51 ddng/gidng liia cao sin vdi 13 ndi vi khuan gay benh bac la Ilia cd doc tinh cao trong khu vuc DBSCL. Bd tri thi nghiem ddi chiing vl so sanh vdi cac gidng liia chuan khing dai dien cho cac gidng mang gien khang vdi tung ddng vi khuan ciia Vien Nghien ciiu Liia Qudc tlGRRI).

a. Dinh gia kieu hinh cic gidng cao sin

Cac gidng liia cao sin ngln ngay dugc gieo trong d c khay khoing dd 15 ngay tudi, sau dd dupe cay ra cac chau trong nha ludi, 15 ngay sau (cay cd khoang tren 10 la) tiln hanh lay nhilm vi khuan.

Song song vdi qui trinh tren vi khuan ciing dugc nuoi va d y chuyin llm sao cho khi din ngly lay nhilm thi sd lupng ciia vi khuan dii dl pha loang dl chiing cho 51 gidng liia. Vi khuan dugc pha din khoang ndng dp 10^^ t l bao/ml. Chuan bi cac dung cu lay nhilm benh nhu: keo (tiet tning), bong thim nude, gim, bao giay bdng md. Chiing ta tiln hanh thu kit qui sau 1 tuIn chiing benh. CIc II dupe thu vl va do de xle dinh kich thude vlt benh. Nlu vlt benh Idn hon 0,5 cm thi dupe coi II nhilm benh.

Gien muc tidu dupe chpn dl thuc hi$n thi nghiem nay la ba gien: Xa -5, Xa-10, Xa-21. Nhiing gien nay cd bilu hien khing dugc benh bac II d ca hai giai doan sinh trudng va phIt triln cua cay liia tren nhilu khu vuc khac nhau (Lang va ctv, 2004).

Gien Xa-4 nlm tren nhilm sic thi sd 11 vl liSn kit chat vdi marker (chi thi) RM 144 va RM 21. Xa-5 nlm tren nhilm sic thi sd 5 va \\ts\ kit vdi marker RG 556, RM 122, RM 13. Cac marker dupe sii dung lam marker dinh dau va dupe diing llm khudn ADN de thilt lap d c cap primer (mdi) dac bi$L Cac cap primer nay se khulch dai cac doan ADN nhd hon nlm trong marker RM 13, RM 122 nhd phuong phIp PCR (Polymerase chaine reaction - phIn ling chudi tning hpp) dugc dinh gil dl chin doln cho gien xa- 5. Gien Xa-10 nlm tren nhilm sic thi sd 11 vdi marker phan tii II RM 206. Cac doan ADN lap lai nay gpi II microsatellite (v€ tinh nhd). Dimg marker RM 122 va cho sin pham khulch dai ADN la 95% chi cd mot bang hinh, khong ghi nhan tien gidng OM 3238. Sin pham khulch dai cho da hinh vdi hai kich tiiudc phan tii khac nhau la 215 bp vdi 240 bp. Cac gidng tuong ling vdi trpng lupng phan tu II240 bp vl tuong ling vdi gien Xa-5 la: OM 4495, OM 2517, OM 2518, OM 2514, OM 5239, OM 5240, OM 2490, OMCS 2000, OM 1490, OM 5636, OM 3408. CIc gidng cdn lai mang gien tuong ling vdi phan hi 215 bp.

Da tim ra 2 chi tiii RM 122 va RM 13 lien kit vdi gien khing Xa-5 tr6n cac gidng liia cao sin la: OM 4495, OM 2517, OM 2518, OM 2514, OM 5239, OM 5240, OM 2490, OMCS 2000, OM 1490, OM 5636, OM 3408, OM 3238, OM 4096, OM 3010, OM 2403, OM 2280. Chin gidng mang gien Xa-21 nhu: OM 4488, OM 5637, OM 4214, OM 4218, OM 4096, OM 6778, OM 5886, OM 3536 vl OM 10011. Tuy nhien gidng Ilia OM 4218 cd hai dang d n chpn Ipc lai.

Gidng OM 4498 mang gien Xa-13, ba gidng mang gien Xa-10 la: OM 4872, DS 2002, OM 4285. Bdn gidng mang gien Xa-2 II: OM 3837, OM 4213, OM 3834, MNR 5; mang gien Xa-1 cd d c gidng: OM 1238, OM 3432, OM 2760, OM 4225, OM 5756; hai gidng mang gien Xa-i^ II: OM 8928 va OM 3243. Bacterial leaf blight caused by the bacterial pathogen Xanthomonas oryzae pv oryzae (Xoo) limits rice yield in all major rice-growing regions of the world, especially in irrigated lowland and rainfed conditions where predisposition factors favor disease development to epidemic proportions. Since bacterial pathogens are difficult to manage, development of host plant resistance is the most effective means of disease management To combat the problem of resistance breakdown, pyramiding of resistance genes into different cultivars is being carried out This study aims at using DNA marker assisted selection (MAS) select varieties that have BBL resistance genes such as Xa-4, Xa-5, Xa-10 and Xa-21. SSR marker with RM 122 and RM 13 linked with xa-5in chromosome 5 such as BB5, OM 4495, OM 2517, OM 2518, OM 2514, OM 5239, OM 5240, OM 2490, OMCS 2000, OM 1490, OM 5636, OM 3408, OM 3238, OM 4096, OM 3010, OM 2403, OM 2280. One STS markers PB7-PB8 deteted genes for Xa-21 such as OM 4488, OM 5637, OM 4214, OM 4218, OM 4096, OM 6778, OM 5886, OM 3536 and OM 10011. Only one xa-131% OM 4498, three varieties bring genes Aa-iCsuch as OM 4872, OMCS 200, OM 2280. For Aa-i" including four new varties OM 3837, OM 4213, OM 3834, MNR 5; gen Xa-1 have five varieties such as OM 1238, OM 3432, OM 2760, OM 4225, OM 5756. For Aa-i^have two varieties OM 8928 and OM 3243. With MNR 1 bring genes Xa-4.

The datta have provided very usefiil information for developing stategies for the development of resistance genes in rice heeding programs to improve varities in Vietnam.
