ABSTRACT
Evaluation of organic matter digestibility and metabolizable energy of ruminant feedstuffs by the in vitro digestion technique with nutrient sources for microbes from rumen fluid
Three experiments were carried out at Cantho University in Mekong delta, Vietnam to evaluate 69 ruminant feedstuffs by the in vitro digestion technique of using rumen fluid (RF) from buffaloes, cattle and goats as nutrient sources for microbes. The results found that the feed organic matter digestibility (OMD) and metabolizable energy (ME) evaluated by the in vitro digestion technique of using 42ml RF plus 8ml buffer solution as medium for microbes (42RF) had a closely linear relationship to those obtained by the In vitro technique standardized by Goering and van Soest (GvS) with the R^> 0.919. The calculated ME value by basing on chemical composition, and OMD of the 42RF at 72h and GvS at 48h were not significantly different (P= 0.878), The conclusions were that the RF from buffaloes, cattle and goats could be used as nutrient sources for microbes to measure the In vitro feed digestibility. The 42RF in vitro technique'had a high potential to evaluate OMD and ME of ruminant feedstuffs.

Robinson va cs, 2004). Ddng thdi nd thudng dugc diing lam nen tang de danh gia chat lugng ciia cac thirc an khac nhau vdi cac ky thuat in vitro hien hiiu (Mabjeesh va cs, 2000;
Al-Masri, 2003; Nguyen Van Thu va Uden, 2003).
Tuy nhien, ky thuat nay van tdn tai nhiing bat lgi do sir dung nhieu loai hoa chat lam dudng chat cho vi sinh vat, ma cac loai hoa chat nay la khan hiem ya tdn kem trong dieu kien nude ta (Nguyen Van Thu, 2006).
Dich dg co (DDC) da dugc biet chiia nhieu dudng chat nhu ammonium, peptide, acid amin, acid beo bay hoi, khoang, vitamin va cac tien td khac dugc hinh thanh tir su phan huy thiirc an va sir tdng hgp ciia vi sinh vat da cd (Kamra, 2005). Cho nen chiing ta cd the tan dung lam mdi trudng dudng chat cho vi sinh vat va ludn san cd d Id md (Nguyen Van Thu, 2006).
Nhiing nghien cim gan day (Danh Mo va Nguyen Van Thu, 2007) da cho thay tiem nang ciia DDC lam ngudn dudng chat cho vi sinh vat de xac dinh ti le tieu hoa thiic an d in vitro.
Do vay, nghien ciiu nay nham muc dich xac dinh kha nang irng dung ky thuat diing DDC ciia Uau, bd va de lam ngudn dudng chat cho vi sinh vat phat trien, de danh gia sir tieu hoa chat hiiu CO va gia tri nang lugng cua thiic an.

Doi tuffng nghiin ciru: Cac gia siic diing de cung cap DDC chi dugc cho an cd tu nhien va dugc thu lay tir Id md.
Bo tri thi nghiem : gdm cd 3 thi nghiem (TN) vdi 69 mau thirc an dugc bd tri nhu sau:
TN 1: cd 32 mau thirc an thudc nhdm cd hg hoa thao, cd hg dau, phu pham, day bau bi, thuy thuc vat va nhdm thirc an khac vdi ngudn DDC sir dung la ciia trau;
TN 2: cd 16 mau thirc an thudc nhdm cd hg boa thao, cd hg dau, phu pham va nhdm thirc an khac vdi ngudn DDC sir dung la cua de
TN 3 : cd 23 mau thirc an thudc nhdm cd hg hoa thao, phu pham trdng trgt, phu pham che bien thuc pham, cd hg dau, banh dau va nhdm thirc an khac vdi ngudn DDC sir dung la ciia bd.

Thanh phin hoa hgc cua cac loai thiic an bao gdm vat chat khd (DM), chat hiru co (OM), dam thd (CP), beo (EE) va xo thd (CF) phan tich theo AOAC (1980), % xo trung tinh (neutral detergent fiber, NDF) va xo axit (acid detergent fiber, ADF) phafi tich theo phuang phap Van Soest vacs. (1991).
Ti le tieu boa chit him co (OMD) in vitro, dugc md ta bdi Goering va van Soest (1970).
Gia tri nang lugng trao ddi (ME), dugc udc tinh theo he thdng ADAS ciia Anh nhu theo (Robinson va cs, 2004) md ta nhu sau:
ME = 0,82 X [0,24 CP + 0,39 EE + 0,18 (OM - CP -EE)] x OMD in vitro.
Gia tri OMD d TNI va TN2 xac dinh d thdi didm 0; 48 va 96 gid va d TN3 xac dinh d thdi didm 0; 24; 48 va 72gid.
Phugng phap nghien cii-u
Danh gia bing 2 ky thuat tieu boa in vitro song song la: 1) Theo dd nghi ciia Goering va van Soest (1970) [GvS]; 2) Bdi ky thuat sir dung 42ml DDC va 8ml dung dich dem (42DDC) thay cac hoa chat trypticase, cac loai khoang da-vi lugng lam dudng chat cho vi sinh vat.
Ky thuat tieu hoa in vitro 42DDC dugc md ta vdi cac budc chinh nhu sau:
Budcl: Can khoang 0,5g mau (mau da dugc nghidn d kich thudc 1mm) [khdi lugng, TLm]
cho vao dng nghiem 175ml
Suae 2: Cho 8ml dung dich dem va 42ml DDC vao dng nghiem da cd mau
Buac 3: Tao ydm khi bing each cho 2ml dung djch oxy hoa khir (reducing) va bom khi CO2 Buac 4: Cac ong nghiem nay dugc ii trong ndi chung each thuy (water bath) d 39-40°C trong khoang thdi gian nhu dien ta phia tren
Budc 5: Cac dng nghiem dugc liy ra khdi ndi chung each thuy, xu ly ngay vdi 50ml dung dich tiy trung tinh (neutral detergent solution) d 85°C trong khoang 12 gid
Bu&c 6- Chuydn toan bd vat chit trong dng nghiem vao cdc lgc (crucible) de rira vdi nude ndng va acetone. Siy cdc lgc d 105°C Uong khoang 12 gid va can (KLl). Tiep tue dem cdc Igc di nung d 500°C trong 2 gid va can (KL2)

Cac budc tiep theo tuong tu nhu ky thuat in vitro 42DDC. Dich da cd dugc lgc qua 3 lap vai muslin trudc khi dimg de thue hien cac thi nghiem.
