I.DATVANDE

2. VAT UEU VA PHUDNG PHAP NGHIBII CUU 2.1. V§t lieu nghien cmi

Phuong phap chon tao dua vao chi thi phan tu va lai tro lai (MABC) duoc sir dung de chpn tao giong Ilia mang da gen chong chiu voi benh bac la, ray nau.

Cac dong khang don tinh trang duoc phat trien bang phuong phap MABC, sau do tiep tuc lai tao cac dong mang da gen khang don tinh trang voi nhau va sang loc kieu hinh, kieu gen de tao ra cac dong mang da gen khang voi da tinh trang (bac la, ray nau) (Hinh 1).

2.2.2 Phuong phap phan ti'ch kieu gen bing chi thi phan tir

ADN duac tach chiet tir mhi la non su dung phuong phap tach nhanh phuc vu cho PCR cua Wang

& cs. (1993). Ti^n hanh ky thuat PCR voi t6ng dung dich phan ung la 15 pi bao gom 50 ng ADN tone so 0,15 pM m6i, 0,2 mM dNTPs, 1 X dich dem PCR 2 5 mM MgCla va 0,5 don vi Taq TaKaRa. h\tM kj-' phan ling PCR: 95"C - 7 phiit; 40 chu ky cua: 940c i s giay, SS^C - 30 giay, 72°C - 2 phiit; giu miu a 4«C S'

2.2.3. Phuong phap danh gia tinh khang/nhiSm benh bac la, ray nau bang lay nhiim nhan tao

- Lay nhiim nhan tao bdnh bac Is. bang phuong phap su dung keo nhiing vao dung dich vi khuan (nong do 10«-10^ CPU) cat dau la lua d€ tao vet thuong. Thi nghiem lay nhiSm duoc thuc hien sau khi cay liia 45 ngay, m6i cay nhiim 5 la vai gi6ng nhilm chuan la 1R24. Ddi chiing am duoc lam vai nuoc cat khu tning. Chieu dai vet benh dugc do sau 18 ngay lay nhiem. Ket qua danh gia dua tren thang danh gia benh tieu chuan (IRRI, 2013).

- Lay nhiim nhan tao ray nau: Ma 10- 15 ngay tuoi (2-3 la) cua gidng nhiem chuan TNI dupc dung de nuoi ray. Ray tuoi 1-2 duoc tha vao long thi nghiem voi mat do 4- 5 con/cay ma. Thai di^m danh gia duoc tien hanh khi 100% giong nhiim chuan TNI ch^t (khoang 10- 15 ngay sau khi tha ray). Ket qua danh gia dua tren hieu chung gay tdn thuong, thang diem dua tren thang danh gia benh tieu chuan (IRRI, 2013).

2.3. Phuong phip xu ly sd li^u

- Du lieu, so heu dong rugng dupc phan tich, xu ly bang cac phan mem IRRISTAT v. 5.0 va Microsoft Excel 2010.

Phan bich sd lieu kieu gen bang phin mem Graphical genotypes 2.0 (GGT v. 2.0).

Bacterial Blight (BB) and Brown Planthopper (BPH) caused by Xantiiomonas oryzae pv. oryzae {Xod) and Nilaparvata lugens Stil, respectively, are the most destructive disease and msect causing significant reduction in nee production. The marker-assisted baclc crossing (MABC) technique is typically used to improve disease and insect resistance for nee. The use of broad-spectrum resistance genes, mtegrating major resistance genes into quality rice varieties, is a strategy to prevent or delay tlie breakdown of resistance of rice cultivars. In this study, multitrait introgression is attempted to combine two desirable traits CBB and BPH resistance) into a genetic background of quality rice variety, Bac Thom 7. The single- trait resistant hues were developed by MABC method, and the crossing among single-trait resistant lines to obtain multitrait introgression lines which earned multiple resistance genes. In each stage of backcrossing, foreground selection was carried out for the target resistance genes (xa5, Xa7, Bph20 snd Bpb21), usmg linked markers, while background selection was done usmg a set of 52 and 56 parental polymorphic SSR markers respectively. The results have been obtained 03 elite lines, N2.3.38.39, N2.3.55.76 and N2.3.64.18, carried 4 resistance genes xa5, Xa7, Bph20anA Bph21, which shown resistance to BB and BPH in artificial infection, and earned the agro-biological characteristics similar to the onginal cultivar, BT7. Line N2.3.38.39, named AGI4, was conducted for author tests, production trials and sent for naUonal basic testing in the spring crop of 2020.
